【 摘 要 】
Theascidianembryo,amodelfortheprimitivemodeofchordatedevelopment,rapidlyformsadorsalnervoussystemwhichconsistsofasmallnumberofneurons.Here,wehavecharacterizedthetranscriptionalregulationofanascidiansynaptotagmin(syt)genetoexplorethemolecularmechanismsunderlyingdevelopmentofsynaptictransmission.Insituhybridizationshowedthatsytisexpressedinallneuronsdescribedinpreviousstudiesandtransientlyintheembryonicepidermis.Neuronalexpressionofsytrequiresinductionfromthevegetalsideoftheembryo,whereasepidermalexpressionoccursautonomouslyinisolatedectodermalblastomeres.Introductionofgreenfluorescentproteinreportergeneconstructsintotheascidianembryosindicatesthatagenomicfragmentofthe3.4-kb5′upstreamregioncontainspromoterelementsofsytgene.Deletionanalysisofthepromotersuggeststhatsytexpressioninneuronsandintheembryonicepidermisdependsondistinctcis-regulatoryregions.【 授权许可】
