【 摘 要 】

Thegenerationofdistinctclassesofmotorneuronsunderliesthedevelopmentofcomplexmotilebehaviorinallanimalsandiswellcharacterizedinchordates.Recentmolecularstudiesindicatethattheascidianlarvalcentralnervoussystem(CNS)exhibitsanteroposteriorregionalizationsimilartothatseeninthevertebrateCNS.Toextendtheunderstandingaboutthediversityofmotorneuronsintheascidianlarva,wehaveidentifiedthenumber,position,andprojectionofindividualmotorneuronsinHalocynthiaroretzi,usingagreenfluorescentproteinunderthecontrolofaneuron-specificpromoter.Threepairsofmotorneurons,eachwithadistinctshapeandinnervationpattern,wereidentifiedalongtheanteroposterioraxisoftheneuraltube:theanteriorandposteriorpairsextendtheiraxonstowarddorsalmusclecells,whereasthemiddlepairprojecttheiraxonstowardventralmuscle.Overexpressionofadominant-negativeformofapotassiumchannelinthesecellsresultedinparalysisontheinjectedside,thusthesecellsmustconstitutethemajorpopulationofmotorneuronsresponsibleforswimmingbehavior.Limclasshomeoboxgeneshavebeenknownascandidategenesthatdeterminesubtypesofmotorneurons.Therefore,theexpressionpatternofHrlim,whichisaLimclasshomeoboxgene,wasexaminedinthemotorneuronprecursors.AllthreemotorneuronsexpressedHrlimatthetailbudstage,althougheachdown-regulatedHrlimatadifferenttime.MisexpressionofHrlimintheepidermallineageledtoectopicexpressionofTuNa2,aputativevoltage-gatedchannelgenenormallyexpressedpredominantlyinthethreepairsofmotorneurons.Hrlimmaycontrolmembraneexcitabilityofmotorneuronsbyregulatingionchannelgeneexpression.
