Pancreaticorganogenesisreliesonacomplexinterplayofcell-autonomousandextracellularsignals.Wedemonstratethatthemorphogensonichedgehog(Shh)isrequiredforpancreaticdevelopmentinzebrafish.GeneticmutantsofShhanditssignalingpathwayestablishthisdependenceasspecifictoendocrine,butnotexocrine,pancreas.Usingcyclopaminetoinhibithedgehogsignaling,weshowthattransientShhsignalingisnecessaryduringgastrulationforsubsequentdifferentiationofendodermintoislettissue.Asecondhedgehog-dependentactivityoccurringlaterindevelopmentwasalsoidentifiedandmaybeanalogoustotheknownactionofShhingutendodermtodirectlocalizationofpancreaticdevelopment.TheearlyactionofShhmaybepartofamoregeneralprocessallowingneuroendocrinecellstooriginateinnonneuroectodermallyderivedtissues.
