【 摘 要 】

Thedirectflightmuscles(DFMs)ofDrosophilaallowforthefinecontrolofwingpositionnecessaryforflight.InDWnt-2mutantflies,certainDFMsareeithermissingorfailtoattachtothecorrectepithelialsites.Usingatemperature-sensitiveallele,weshowthatDWnt-2activityisrequiredonlyduringpupationforcorrectDFMpatterning.DWnt-2isexpressedintheepitheliumofthewinghingeprimordiumduringpupation.ThisexpressionisinthevicinityofthedevelopingDFMs,asrevealedbyexpressionofthemusclefoundercell-specificgenedumbfoundedinDFMprecursors.TheobservationthatagenenecessaryforembryonicfoundercellfunctionisexpressedintheDFMprecursorssuggeststhatthesecellsmayhaveasimilarfoundercellrole.AlthoughtheexpressionpatternofDWnt-2suggeststhatitcouldinfluenceepithelialcellstodifferentiateintoattachmentsitesformuscle,theexpressionofstripe,atranscriptionfactornecessaryforepithelialcellstoadoptanattachmentcellfate,isunalteredinthemutant.EctopicexpressionofDWnt-2inthewinghingeduringpupationcanalsocreatedefectsinmusclepatterningwithoutalterationsinstripeexpression.WeconcludethatDWnt-2promotesthecorrectpatterningofDFMsthroughamechanismthatisindependentoftheattachmentsitedifferentiationinitiatedbystripe.
