【 摘 要 】

Nodalsignallingisessentialformanydevelopmentaleventsduringvertebratedevelopment,includingtheestablishmentofleft–rightasymmetry,ofdorsoventralaxisofthecentralnervoussystem,andendodermandmesodermformation.ThezebrafishTGFβ-relatedtypeIreceptor,TARAM-A(Tar),isexpressedintheprospectivemesendodermalterritoryand,whenactivated,cantransfateearlyblastomeresintoendoderm,suggestingthatNodalandTarmayrepresentsimilarsignallingpathways.Wehaveanalysedthefunctionalrelationshipsbetweenthosetwopathwaysinzebrafish.Wefirstdemonstratethattarandthezebrafishnodalgenescycandsqtfunctionallyinteract.Wealsoshowthatadominant-negativeisoformofTar,TarMR,interferesspecificallywiththefunctionofCycandSqtinvitro,butdoesnotinterferewiththefunctionofBMP2,anotherTGFβ-relatedmolecule.TarMRinterferesalsowithNodalsignallinginvivosinceitenhancesthephenotypeofembryoswithweakenedNodalsignalling.OverexpressionoftarMRinwild-typeembryosinterferedwiththeformationofendoderm-derivedstructures.Conversely,overexpressionoftarenlargedthepresumptivemesendodermalregionattheonsetofgastrulation.Together,ourresultspointtoTarasanessentialfactorforendodermformationandanimportantmodulatorofNodalsignalling,potentiallyrepresentingoneoftheNodalreceptors.
