【 摘 要 】

Atthelateblastocyststage,theepithelialtrophectodermcellsofthemammalianembryoundergoaphenotypicchangethatallowsthemtoinvadeintotheuterinestromaandmakecontactwiththematernalcirculation.Thisstepcanberegulatedinvitrobytheavailabilityofaminoacids.Embryosculturedindefinedmediumlackingaminoacidscannotformtrophoblastcelloutgrowthsonfibronectin,aninvitromodelofimplantation,butremainviableforupto3daysincultureandwillformoutgrowthswhentransferredintocompletemedium.Theaminoacidrequirementisadevelopmentallyregulatedpermissiveeventthatoccursduringa4-to8-hperiodattheearlyblastocyststage.Aminoacidsaffectspreadingcompetencespecificallybyregulatingtheonsetofprotrusiveactivityandnottheonsetofintegrinactivation.Rapamycin,aspecificinhibitorofthekinasemTOR/FRAP/RAFT1,blocksaminoacidstimulationofembryooutgrowth,demonstratingthatmTORisrequiredfortheinitiationoftrophectodermprotrusiveactivity.Inhibitionofglobalproteintranslationwithcycloheximidealsoinhibitsaminoacid-dependentsignals,suggestingthatmTORregulatesthetranslationofproteinsrequiredfortrophoblastdifferentiation.OurdatasuggestthatmTORactivityhasadevelopmentalregulatoryfunctionintrophectodermdifferentiationthatmayservetocoordinateembryoanduterusatthetimeofimplantation.
