【 摘 要 】
Forendocyticuptakeofvitellogenins,developingoocytesofOncopeltusrequireasoluble,diffusiblemolecularsignalfromtheirsurroundingepithelialcells,andthissignalmustbetransmittedthroughopengapjunctions.Hormonalstimulationtriggeringsynthesisandprocessingofvitellogeninsintomatureinsectyolksphereshasbeenintenselystudied,andfollicleepithelialcellsareknowninseveralinsectstocontributetothebloodproductswhichareendocytosedalongwithvitellogeninssynthesizedinthefatbodies.However,therehasbeenlittleevidencethatdirectgapjunctionalcommunicationisarequirementforendocyticactivitybyoocytes.Inuntreatedcontrolfollicles,bothelectricalanddyecouplingoccur,andfolliclesincubatedinvitroinphysiologicalsaltsolutioncontainingsmallamountsofbloodandfluorescentdyeproducefluorescentlylabelednascentyolkspheres.Labeledyolksphereswerevisibleinbothsectionedmaterial,and,with(Laser)ConfocalScanning,inlivingmaterial.Dyecouplingwasabolishedbytreatmentwitheither1mMoctanol,0.5mMethylmethanesulfonate(EMS),orcytoplasmicacidification,withcouplingcoefficientsalsobeingaffectedaseachofthesegapjunctionantagonistsdown-regulatedtheconnexons.Witheachofthesetreatments,aftergapjunctionsweredown-regulated,receptor-mediatedendocyticuptakeofblood-bornvitellogeninscametoahalt.Furthermore,Oncopeltusfollicleswithendocyticactivityblockedinthismannercouldberescuedbymicroinjectionofthesolublefractionoflysedepithelialcellcytoplasm,confirmingthattheprocessdependeduponamolecularsignalfromtheepithelialcells.
