【 摘 要 】

Interactionsbetweeningrowingnervefibersandtheirtargettissuesformthebasisforfunctionalconnectivitywiththecentralnervoussystem.Studiesofthedevelopingdentalpulpinnervationbynervefibersfromthetrigeminalganglionisanexcellentexampleofnerve–targettissueinteractionsandwillallowspecificquestionsregardingdevelopmentofthedentalpulpnervesystemtobeaddressed.Dentalpulpcells(DPC)produceanarrayofneurotrophicfactorsduringdevelopment,suggestingthattheseproteinsmightbeinvolvedinsupportingtrigeminalnervefibersthatinnervatethedentalpulp.Wehaveestablishedaninvitroculturesystemtostudytheinteractionsbetweenthedentalpulpcellsandtrigeminalneurons.Weshowthatdentalpulpcellsproduceseveralneurotrophicfactorsinculture.WhenDPCarecoculturedwithtrigeminalneurons,theypromotesurvivalandaspecificandelaborateneuriteoutgrowthpatternfromtrigeminalneurons,whereasskinfibroblastsdonotprovideasimilarsupport.Inaddition,weshowthatdentalpulptissuebecomesinnervatedwhentransplantedectopicallyintotheanteriorchamberoftheeyeinrats,andupregulatesthecatecholaminergicnervefiberdensityoftheirises.Interestingly,graftingthedentalpulptissueintohemisectedspinalcordincreasesthenumberofsurvivingmotoneurons,indicatingafunctionalbioactivityofthedentalpulp-derivedneurotrophicfactorsinvivobyrescuingmotoneurons.Basedonthesefindings,weproposethatdentalpulp-derivedneurotrophicfactorsplayanimportantroleinorchestratingthedentalpulpinnervation.
