TheprevailingconcepthasbeenthatanFGFinducesepithelial-to-fiberdifferentiationinthemammalianlens,whereaschicklenscellsareunresponsivetoFGFandareinsteadinducedtodifferentiatebyIGF/insulin-typefactors.Weshowherethatwhentreatedforperiodsinexcessofthoseusedinpreviousinvestigations(>5h),purifiedrecombinantFGFsstimulateproliferationofprimaryculturesofembryonicchicklensepithelialcellsand(athigherconcentrations)expressionofthefiberdifferentiationmarkersδ-crystallinandCP49.Surprisingly,upregulationofproliferationandδ-crystallinsynthesisbyFGFdoesnotrequireactivationofERKkinases.ERKfunctionis,however,essentialforstimulationofδ-crystallinexpressioninresponsetoinsulinorIGF-1.Vitreoushumor,thepresumptivesourceofdifferentiation-promotingactivityinvivo,containsafactorcapableofdiffusingoutofthevitreousbodyandinducingδ-crystallinandCP49expressioninchicklenscultures.Thisfactorbindsheparinwithhighaffinityandincreasesδ-crystallinexpressioninanERK-insensitivemanner,propertiesconsistentwithanFGFbutnotinsulinorIGF.OurfindingsindicatethatdifferentiationinthechicklensislikelytobemediatedbyanFGFandprovidethefirstinsightsintotheroleoftheERKpathwayingrowthfactor-inducedsignaltransductioninthelens.
