TheproteintyrosinephosphatasePTP1,whichmediatesreversiblephosphorylationontyrosine,hasbeenshowntoplayanimportantregulatoryroleduringDictyosteliumdevelopment.MutantslackingPTP1developmorerapidlythannormal,whilestrainsthatoverexpressPTP1displayaberrantmorphology.However,thesignallingpathwaysinvolvedhavenotbeencharacterised.Inreexaminingthesestrains,wehavefoundthatthereisaninversecorrelationbetweenlevelsofPTP1activity,theextentoftyrosinephosphorylationonDictyosteliumSTATaaftertreatmentwithcAMP,andtheproportionoftheslugpopulationexhibitingSTATanuclearenrichmentinvivo.ThissuggeststhatPTP1actstoattenuatethetyrosinephosphorylationofSTATaanddownstreamSTATa-mediatedpathways.Consistentwiththis,weshowthatwhenPTP1isoverexpressed,thereisincreasedexpressionofaprestalkcellmarkerattheslugposterior,aphenocopyofSTATanullslugs.Inptp1nullstrains,STATatyrosinephosphorylationandnuclearenrichmentinthesluganteriorisincreased.Thereisalsoachangeintheprestalktopresporecellratio.Synergyexperimentssuggestthatthisisduetoacell-autonomousdefectinformingthesubsetofpresporecellsthatarelocatedintheanteriorpresporeregion.
