【 摘 要 】
StudiesfromourgroupandothershaveshownthatinvitrosomitemyogenesisisregulatedbyneuraltubeandnotochordfactorsincludingWnt,Sonichedgehog(Shh),andbasicfibroblastgrowthfactor(bFGF)togetherwithtransforminggrowthfactor-β1(TGF-β1).Inthisstudywereportthatinsulinandinsulin-likegrowthfactorsIandII(IGF-Iand-II)alsopromotemyogenesisinexplantculturescontainingsinglesomitesorsomite-sizedpiecesofsegmentalplatemesodermfrom2-day(stage10–14)chickenembryosandthatthecombinationofinsulin/IGFswithbFGFplusTGF-β1promotesevenhigherlevelsofmyogenesis.WealsofoundthatShhpromotesmyogenesisinthisinvitrosystemandthatShhinteractssynergisticallywithinsulin/IGFstopromotehighlevelsofmyogenesis.RT-PCRanalysisdetectedinsulin,IGF-II,insulinreceptor,andIGFreceptormRNAsinboththeneuraltubeandthesomites,whereasIGF-Itranscriptsweredetectedinentireembryosbutnotintheneuraltubeorsomites.Treatmentofsomite–neuraltubecocultureswithanti-insulin,anti-IGF-II,anti-insulinreceptor,oranti-IGFreceptorblockingantibodiescausedasignificantdecreaseinmyogenesis.TheseresultsareconsistentwiththehypothesisthatsystemicIGF-IaswellasinsulinandIGF-IIsecretedbytheneuraltubeactasadditionalearlymyogenicsignalsduringembryogenesis.Furtherstudiesindicatethatinsulin,IGFs,bFGF,andShhalsostimulatesomitecellproliferationandinfluenceapoptosis.
