Interactionsbetweenepithelialandmesenchymaltissuesinthedevelopinginnereardirecttheformationofitscartilaginouscapsule.Recentworkindicatesthatmanygrowthfactorsaredistributedintheearlyembryoinvivoinatemporal-spatialpatternthatcorrelateswithsitesofongoingmorphogeneticevents.Wereportherethelocalizationoftransforminggrowthfactorβ1(TGF-β1inbothepithelialandmesenchymaltissuesofthemouseinnerearbetween10and16daysofembryonicdevelopment(E10–E16).Inaddition,utilizingahigh-densityculturesystemasaninvitromodelofoticcapsulechondrogenesis,weshowthatmodulationofchondrogenesisbyTGF-β1inculturedmouseperioticmesenchymemimicstheinvitroeffectsofoticepitheliumontheexpressionofchondrogenicpotential.WeprovideevidenceofacausalrelationshipofthisgrowthfactortooticcapsuleformationinsitubydemonstratingthattheactualsequenceofchondrogeniceventsthatoccurinthedevelopingembryoisreproducedinculturebytheadditionofexogenousTGF-β1peptide.Furthermore,inculturesofmesenchymecontainingoticepithelium,wedemonstratethelocalizationofendogenousTGF-β1,firstwithintheepithelialtissueandlaterwithinboththeepitheliumanditssurroundingperioticmesenchyme,contrastedtoanabsenceofendogenousTGF-β1inculturesofmesenchymealone.OurresultssuggestthatTGF-β1isoneofthesignalmoleculesthatmediatetheeffectsofoticepitheliumininfluencingtheformationofthecartilaginousoticcapsule
