【 摘 要 】

Fibronectinhasbeenshowntoenhancecellmigrationinvitro.Itseemsofconsiderableinterest,therefore,tomapitsdistributionintheactualembryoduringperiodsofcellmigration.Inthisinsitustudy,weusedimmunohistochemistryattheelectronmicroscopicleveltolocalizefibronectininthreeregionsofthechickembryothatarecharacterizedbyactualorimpendingmorphogeneticmovements:dorsaltrunk(neuralcrest),ventraltrunk,andareavasculosa.Theextracellularmatrixofthe2-day-oldembryocontainsnonstriatedfibrils(5–10nmindiameter)andso-calledinterstitialbodies,bothofwhichlabelbyantibodiestofibronectinwiththeimmunoperoxidase(PAP)techniqueandwiththeimmunoferritintechnique.Inaddition,cellsurfacesandbasallaminaearestrainedbyPAPfollowingtreatmentwithantifibronectin.Usingrutheniumredtopreserveproteoglycan(PG),wewereabletodemonstrateacloseassociationofPGgranuleswiththeperipheryofthefibronectin-richinterstitialbodies.Theseresultsarethefirsttoindicatethepossiblechemicalcompositionofinterstitialbodiesandtheyraisethepossibilitythattheseuniquecomponentsofembryonicmatricesareimportantsubstratesforcellmigration.Indeed,cellprocessesofneuralcrestandothermesenchymemakeclosecontactswithinterstitialbodies.Intheareavasculosa,interstitialbodiesareconcentratedattheleadingedgeofthevascularmesenchyme.Thereis,however,noobviousgradientofextracellularfibronectininneuralcrestpathwaysorintheventraltrunk.
