Detection of Potato virus Y (PVY) in potato tubers in Antioquia (Colombia) using TAS-ELISA and qRT-PCR

【 摘 要 】

PVY is one of the viruses with highest incidence and severity in potato crops in Colombia and other countries. This virus is transmitted by aphids and tuber seeds and can progressively reduce crop yields due to viral accumulation when non-certified seed is used. In this work, the incidence and distribution of PVY in tubers of Solanum tuberosum v. Diacol-Capiro and S. phureja v. Criolla-Colombia from Antioquia (Colombia) was evaluated using TAS-ELISA and qRT-PCR. For both varieties, 16 samples from four different tissues (skin, dormant buds, base and tip of sprouts) were used and detection levels were compared for both techniques. With qRT-PCR, PVY was detected in 100 % and 66.6 % of Diacol-Capiro and Criolla-Colombia tubers, respectively. In contrast, TAS-ELISA detected PVY in only 75 % of Diacol-Capiro samples and none of Criolla-Colombia. Low titer values may explain the failure to detect PVY by TAS-ELISA in some samples that tested positive by qRT-PCR in Criolla-Colombia (Ct=32.4) and Diacol-Capiro (Ct=15.54). Distribution analysis revealed that the highest accumulation of PVY was the tip region of sprouts in both varieties (Diacol-Capiro Ct=11.53 and Criolla-Colombia Ct=27.52). These findings demonstrate the need of using certified seed-tubers tested with highly sensitive methods like qRT-PCR.
