【 摘 要 】
Chlamydiatrachomatisisanobligateintracellularbacteriumresponsibleforoneofthemostcommonsexuallytransmitteddiseases.Inepithelialcells,C.trachomatisresidesinamodifiedmembrane‐boundvacuoleknownasaninclusion,whichisisolatedfromtheendocyticpathway.However,thematurationprocessofC.trachomatiswithinimmunecells,suchasmacrophages,hasnotbeenstudiedextensively.Here,wedemonstratedthatRAWmacrophageseffectivelysuppressedC.trachomatisgrowthandpreventedGolgistackdisruption,ahallmarkdefectinepithelialcellsafterC.trachomatisinfection.Next,wesystematicallyexaminedassociationbetweenC.trachomatisandvariousendocyticpathwaymarkers.Spinningdiskconfocaltime‐lapsestudiesrevealedsignificantandrapidassociationbetweenC.trachomatiswithRab7andLAMP1,markersoflateendosomesandlysosomes.Moreover,pretreatmentwithaninhibitoroflysosomeacidificationledtosignificantincreasesinC.trachomatisgrowthinmacrophages.Atlaterstagesofinfection,C.trachomatisassociatedwiththeautophagymarkerLC3.TEManalysisconfirmedthatasignificantportionofC.trachomatisresidedwithindouble‐membrane‐boundcompartments,characteristicofautophagosomes.Together,theseresultssuggestthatmacrophagescansuppressC.trachomatisgrowthbytargetingitrapidlytolysosomes;moreover,autophagyisactivatedatlaterstagesofinfectionandtargetssignificantnumbersoftheinvadingbacteria,whichmayenhancesubsequentchlamydialantigenpresentation...
