CD1d‐restrictedtypeINKTcellsprovidehelpforspecificantibodyproduction.Bcells,whichhavecapturedandpresentedaT‐dependent,antigen‐derivedpeptideonMHCclassIIandCD1d‐bindingglycolipidα‐GConCD1d,respectively,activateThandNKTcellstoelicitBcellhelp.However,theroleoftheDCCD1dinhumoralimmunityremainsunknown.WethereforeconstructedmixedbonemarrowchimerascontainingCD1d‐expressing,DTR‐transgenicDCsandCD1d+orCD1d−nontransgenicDCs.FollowingDT‐mediatedDCablationandimmunization,weobservedthattheprimaryandsecondaryantibodyresponseswereequivalentinthepresenceofCD1d+andCD1d−DCs.Incontrast,atotalablationofDCsdelayedtheprimaryantibodyresponse.FurtherexperimentsrevealedthatdepletionofCD1d+DCsblockedinvivoexpansionofantigen‐specificcytotoxic(CD8+)Tlymphocytes.TheseresultsprovideacleardemonstrationthatalthoughCD1dexpressiononDCsisessentialforNKT‐enhancedCD8+Tcellexpansion,itisdispensableforspecificantibodyproduction...
