Bacterialandviralrespiratorytractinfectionsresultinmillionsofdeathsworldwideandarecurrentlytheleadingcauseofdeathfrominfection.Acuteinflammationisanessentialelementofhostdefenseagainstinfection,butcanbedamagingtothehostwhenleftunchecked.Effectivehostdefenserequiresmultiplelipidmediators,whichcollectivelyhaveproinflammatoryand/orproresolvingeffectsonthelung.Duringpulmonaryinfections,phospholipidacylchainsandcholesterolcanbechemicallyandenzymaticallyoxidized,aswellastruncatedandmodified,producingcomplexmixturesofbioactivelipids.Wereviewrecentevidencethatphospholipidsandcholesterolandtheirderivativesregulatepulmonaryinnateandadaptiveimmunityduringinfection.Wefirsthighlightdatathatoxidizedphospholipidsgeneratedinthelungduringinfectionstimulatepatternrecognitionreceptors,suchasTLRsandscavengerreceptors,therebyamplifyingthepulmonaryinflammatoryresponse.Next,wediscussevidencethatoxidationofendogenouspoolsofcholesterolduringpulmonaryinfectionsproducesoxysterolsthatalsomodifythefunctionofbothinnateandadaptiveimmunecells.Last,weconcludewithdatathatn‐3polyunsaturatedfattyacids,bothintheformofphospholipidacylchainsandthroughenzymaticprocessingintoendogenousproresolvinglipidmediators,aidintheresolutionoflunginflammationthroughdistinctmechanisms.Unravelingthecomplexmechanismsofinductionandfunctionofdistinctclassesofbioactivelipids,bothnativeandmodified,mayholdpromisefordevelopingnewtherapeuticstrategiesforimprovingpulmonaryoutcomesinresponsetoinfection...
