Bothα‐andβ‐defensinshaveanti–humanimmunodeficiencyvirusactivity.Thesedefensinsachievehumanimmunodeficiencyvirusinhibitionthroughavarietyofmechanisms,includingdirectbindingwithvirions,bindingtoandmodulationofhostcell‐surfacereceptorswithdisruptionofintracellularsignaling,andfunctioningaschemokinesorcytokinestoaugmentandalteradaptiveimmuneresponses.Polymorphismsinthedefensingeneshavebeenassociatedwithsusceptibilitytohumanimmunodeficiencyvirusinfectionanddiseaseprogression.However,therolesthatthesedefensinsandtheirgeneticpolymorphismshaveininfluencinghumanimmunodeficiencyvirus/acquiredimmunodeficiencysyndromeoutcomesarenotstraightforwardand,attimes,appearcontradictory.Differencesinpopulations,studydesigns,andtechniquesforgenotypingdefensingenepolymorphismsmayhavecontributedtothislackofclarity.Inaddition,acomprehensiveapproach,wherebothsubfamiliesofdefensinsandtheirall‐inclusivegeneticpolymorphismprofilesareanalyzed,islacking.Suchanapproachmayrevealwhetherthehumanimmunodeficiencyvirusinhibitoryactivitiesofα‐andβ‐defensinsarebasedonparallelordivergentmechanismsandmayprovidefurtherinsightsintohowthegeneticpredispositionforsusceptibilityorresistancetohumanimmunodeficiencyvirus/acquiredimmunodeficiencysyndromeisorchestratedbetweenthesemolecules...
