【 摘 要 】
MammalianIDOisaheme‐containingenzymewhosemainactivityinmammalsistodegradetheessentialaminoacidtrypintol‐kynurenine.Althoughthelinkbetweenitsenzymaticactivityandtheimmuneresponseisnotstraightforward,severallinesofevidencesuggestthatthisenzymeisinvolvedinfightinginfectionsandparadoxically,alsointheestablishmentoftheimmunetoleranceassociatedwithfetusimplantationandwiththedevelopmentofoncogenicprocesses.IDOisassociatedwiththesuccessfuldevelopmentofthefetus.Itparticipatesearlyinpregnancytotheefficientinvasionoftheuterinemucosabythenascenttrophoblastandremainsactivethroughoutthewholeprocess,asillustratedbythedecreaseinsystemictrypfromthesecondtrimesterofgestationandthereturntonormalvaluesafterdelivery.Theshort‐termactivationofIDOinresponsetoinvadingpathogensandemergingtumorsparticipatesintheeliminationofthesethreats,whereasthesustainedactivationofIDOoftenresultsinastateofimmunetolerancethatmayfavorchronicinfectionsandtheuncontrolledproliferationofmalignantcells.However,despitethesepotentialdeleteriouseffectsofIDO,theenzymeisinstrumentalinmaintainingtheperipheraltolerancethatisrequiredtoavoidautoimmunediseases.Below,wereviewtheimplicationofIDOactivationuponthephysiologicaldevelopmentofthefetusandthepathologicaldevelopmentoftumorsanddiscusswhethersuchanenzymecouldbeusedasatherapeutictooltodecreasetherateofallograftrejectionsviaitspotentimmunomodulatoryproperties...
