Inthepresentstudies,wehaveestablishedthatRSVcanelicitamorepathogenicenvironmentdependentonimproperDC‐associatedsensitization.OurinitialstudiesdemonstratedthatRSV,butnotinfluenza,infectionduringanallergenexposureintotheairwayinducedamoresevereallergenresponse.TheRSV‐inducedexacerbationincludedanincreasedTh2cytokineresponseandpathophysiologyasmonitoredbyAHRandmucusoverproduction.DCsplayedacentralroleintheallergen‐inducedresponses,asinstillingRSV‐infectedBMDCintotheairwaycouldrecapitulatealiveviruschallenge.WiththeuseofCCR6−/−micethathaveaprimarydefectintherecruitmentofmDCsubsets,reducedexacerbationofdiseasewasobservedwhenRSVwasadministeredalongwithallergen.Furthermore,sensitizationofmicewithRSV‐infectedBMDCintotheairwayproducedamoresevereimmuneresponsetoaliveviruschallenge.Subsequently,usingRSV‐infectedBMDCfromCCR7−/−mice(thatdonotmigrateefficientlytoLNs)tosensitizetheexacerbatedresponsedemonstratedthattheresponsewasdependentonDCmigrationtotheLN.Finally,theabilityofRSV‐infectedDCstoelicitanexacerbated,allergen‐inducedpathogenicresponsecouldbemaintainedforaslongas3weeks,suggestingthatRSV‐infectedDCsthemselvescreatedanalteredimmuneenvironmentthatimpactsoff‐targetmucosalresponsesthatcouldhaveprolongedeffects...
