STAT1‐dependent IgG cell‐surface expression in a human B cell line derived from a STAT1‐deficient patient

【 摘 要 】

STAT1isakeyeffectorofcytokinesinvolvedintheresistancetopathogens;itsidentifiedtranscriptionaltargetsmediatetheinnateimmuneresponseinvolvedinthedefenseagainstvirusesandbacteria.LittleisknownabouttheroleofSTAT1inadaptiveimmunity,includingitsimpactonBCRorsurfaceIgexpression.Analysisofthispointisdifficultinhumans,asSTAT1deficiencyisextremelyrare.SDpatientsdieearlyinchildhoodfromasevereimmunodeficiency.Herein,aSDBcelllineobtainedfromaSDpatientwascomparedwithaBcelllinefromaSTAT1‐proficientsubjectinsearchofdifferencesinsurfaceIgexpression.InthisSDBcellline,acompleteabsenceofsurfaceIgGwasnoted.ThemRNAencodingthesurfaceformofIgGwasdetectedonlyinSTAT1‐proficientBcells;themRNAsencodingthesecretedandthesurfaceformsweredetectedinSDandSTAT1‐proficientBcells.Re‐expressionofSTAT1inSDBcellsrestoredsurfaceIgGexpressionandafunctionalBCR.Conversely,shRNAsilencingofSTAT1inBcellsreducedconsiderablytheexpressionofthesurfaceIgG.AlthoughlimitedtooneBcellline,theseresultssuggestthatSTAT1mayplayanessentialroleinsurfaceIgGexpressioninhumanBcells.PossiblemechanismsinvolveregulationofmRNAsplicing,transcription,orboth.TheseobservationsextendtheroleofSTAT1furtherinadaptiveimmunity,includingtheregulationofBCRexpression...
