【 摘 要 】
HumanMSCsmayrespondtoTLRligation,andrecentresearchhassuggestedthatmanytissuescontaintissue‐specificMSCs,possiblylocatedinperiendothelialandperivascularregions.Atpresent,thefunctionalconsequencesofthesefindingsareunclear.Wehypothesizedthattissue‐specificMSCscouldplayaninstructionalroleduringearlyphasesofbacterialchallenge.Toinvestigatethishypothesisfurther,wesetupacoculturesystemofglandularMSCsandperipheralbloodneutrophilssothatwecouldanalyzethecellularinteractionsofthesecellsinresponsetoLPSchallenge.WefoundthatstimulationwithbacterialendotoxininducedchemokinereceptorexpressionandmobilityofMSCs.ActivatedMSCssecretedlargeamountsofinflammatorycytokinesandrecruitedneutrophilsinanIL‐8‐andMIF‐dependentmanner.Recruitedandactivatedneutrophilsshowedaprolongedlifespan,anincreasedexpressionofinflammatorychemokines,andanenhancedresponsivenesstowardsubsequentchallengewithLPS.Ourfindingsdemonstrateacomplex,functionalinteractionbetweentissue‐residentMSCsandperipheralbloodneutrophilsuponbacterialchallengeandsuggestaroleforMSCsintheearlyphasesofpathogenchallenge,whenclassicalimmunecellshavenotbeenrecruitedyet...
