Myeloid‐derived suppressor cells in the peripheral blood of cancer patients contain a subset of immature neutrophils with impaired migratory properties

Intumor‐bearingmice,immunosuppressivegranulocyticandmonocyticMDSChavebeenidentified.TheidentityandfunctionofMDSCincancerpatientsarelessclearandneedfurthercharacterization.Weanalyzedtheperipheralbloodof103patientswithHNC,lungcancer,orcancersofbladderandureter.Basedonsedimentationpropertiesindensitygradients,asubsetofLD‐PMNwasidentifiedandanalyzed.LD‐PMNwereexpandedintheperipheralbloodofcancerpatients,suppressedproliferation,andIFN‐γproductionofpolyclonallystimulatedTcellsandthus,qualifyashumanMDSC.ImmunophenotypingandmorphologicalanalysisrevealedtheaccumulationofimmaturePMNintheMDSCfraction.NeutrophilicMDSCshowedalteredsurfacemarkerexpression,prolongedsurvival,andimpairedeffectorfunctionswhencomparedwithconventional,maturePMNofregulardensity.MDSCdisplayedmarkedlyreducedchemotaxistowardtumor‐conditionedmediumandlackedexpressionofchemokinereceptorsCXCR1andCXCR2,whicharenormallyrequiredforPMNextravasationfromthebloodstreamandsubsequenttissueinfiltration.Collectively,ourdatasuggesttheaccumulationandpersistenceoflong‐lived,immaturegranulocyticMDSCwithTcell‐suppressivefunctionandimpairedmigratorypropertiesintheperipheralbloodofcancerpatients...
