Thelungisconstantlyexposedtopotentiallypathogenicparticlesandmicroorganisms.Ithasbecomeevidentrecentlythatnotonlyinnatebutalsoadaptiveimmuneresponsestoparticulates,suchasSiO2enteringtherespiratorytract,arecomplexanddynamicevents.Althoughthecellularmechanismsandanatomicalconsequencesinvolvedinthedevelopmentofsilicosishavebeenstudiedextensively,theystillremainpoorlyunderstood.Basedontheircapacityforimmuneregulation,lymphocytesmayplayakeyroleintherespiratoryresponsetoenvironmentalchallengebySiO2.TheobjectiveofthisstudywastocharacterizetheimpactofSiO2exposureonrespiratoryimmuneprocesses,withparticularemphasisonevaluatingtheimportanceoflymphocytesinthemurinesilicosismodel.Therefore,lymphopenicmice,includingNK‐deficient,Rag1−/−,oracombination(Rag1−/−NK‐depleted),wereusedanddemonstratedthatSiO2‐inducedfibrosisandinflammationcanoccurindependentlyofT,B,NKT,andNKcells.StudiesinRag1−/−micesuggestfurtherthatlymphocytesmayparticipateintheregulationofSiO2‐inducedinflammationthroughmodulationoftheNalp3inflammasome.Thisobservationmayhaveclinicalrelevanceinthetreatmentofinflammatoryandfibroticlungdiseasesthatarerefractoryorrespondsuboptimallytocurrenttherapeutics...
