TheimmunostimulatoryroleofKupffercellsinvariousinflammatoryliverdiseasesisstillnotfullyunderstood.Inthisstudy,phenotypicandfunctionalaspectsofKupffercellsfromhealthyC57BL/6micewereanalyzedandcomparedwiththoseofsplenicandperitonealmacrophagestogenerateablueprintofthecellsundersteady‐stateconditions.Inthemouseliver,onlyonepopulationofKupffercellswasidentifiedasF4/80highCD11blowcells.WeobservedthatfreshyisolatedKupffercellsareendocyticandshowarelativelyhighbasalROScontent.Interestingly,despiteexpressionofTLRmRNAonKupffercells,ligationofTLR4,TLR7/8,andTLR9resultedinaweakinductionofIL‐10,loworundetectablelevelsofIL‐12p40andTNF,andup‐regulationofCD40onthesurface.Kupffercellsandsplenicmacrophagesshowfunctionalsimilarities,incomparisonwithperitonealmacrophages,asreflectedbycomparablelevelsofTLR4,TLR7/8,andTLR9mRNAandloworundetectablelevelsofTNFandIL‐12p40produceduponTLRligation.Theunique,functionalcharacteristicsofKupffercells,demonstratedinthisstudy,suggestthatKupffercellsundersteady‐stateconditionsarespecializedasphagocytestoclearanddegradeparticulatesandonlyplayalimitedimmunoregulatoryroleviathereleaseofsolublemediators...
