【 摘 要 】
Neutrophilsarethefirstlineofdefenseafterapathogenhasbreachedtheepithelialbarriers,andunimpairedneutrophilfunctionsareessentialtoclearinfections.Staphylococcusaureusisaprevalenthumanpathogenthatisabletowithstandneutrophilkilling,yetthemechanismsusedbyS.aureustoinhibitneutrophilclearanceremainincompletelydefined.Theproductionofreactiveoxygenspecies(ROS)isavitalneutrophilantimicrobialmechanism.Herein,wetestthehypothesisthatS.aureususestheSaeR/Stwo‐componentgeneregulatorysystemtoproducevirulencefactorsthatreduceneutrophilROSproduction.WiththeuseofROSprobes,thetemporalandoverallproductionofneutrophilROSwasassessedduringexposuretotheclinicallyrelevantS.aureusUSA300(strainLAC)anditsisogenicmutantLACΔsaeR/S.OurresultsdemonstratedthatSaeR/S‐regulatedfactorsdonotinhibitneutrophilsuperoxide(O2−)production.However,subsequentneutrophilROSproductionwassignificantlyreducedduringexposuretoLACcomparedwithLACΔsaeR/S.Inaddition,neutrophilH2O2productionwasreducedsignificantlybySaeR/S‐regulatedfactorsbyamechanismindependentofcatalase.Consequently,thereductioninneutrophilH2O2resultedindecreasedproductionofthehighlyantimicrobialagenthypochlorousacid/hypochloriteanion(HOCl/−OCl).ThesefindingssuggestanewevasionstrategyusedbyS.aureustodiminishavitalneutrophilantimicrobialmechanism...
