Characterization of lung infection–induced TCRγδ T cell phenotypes by CyTOF mass cytometry

TcellreceptorγδcellsareknowntobetheprimaryeffectorTcellsinvolvedintheresponsetobacterialinfections,yettheirphenotypiccharacteristicsarenotaswellestablishedasotherTcellsubsets.Inthisstudy,weusedcytometrybytime‐of‐flightmasscytometrytobettercharacterizethephenotypicresponseofTcellreceptorγδcellstoStreptococcuspneumoniaelunginfection.Micewereinfected,andcellsfromlungwashouts,spleen,andlymphnodeswerestainedtodetectcell‐surface,intracellular,andsignalingmarkers.WeobservedthatinfectioncausedasignificantincreaseinTcellreceptorγδcells,whichexpressedhighinterferon‐γandinterleukin‐17Alevels.ProfilingTcellreceptorγδcellsbycytometrybytime‐of‐flightrevealedthatactivatedγδTcellsuniquelycoexpressedcell‐surfaceGr‐1,clusterofdifferentiation14,andclusterofdifferentiation274(programmeddeath‐ligand1).FurtherclassificationofGr‐1expressionpatternsonTcellreceptorγδcellsdemonstratedthatGr‐1+Tcellreceptorγδcellsweretheprimarysourceofinterferon‐γ,whereasGr‐1−cellsmostlyexpressedinterleukin‐17A.Gr‐1+Tcellreceptorγδcellsalsoshowedhigherζ‐chain–associatedproteinkinase70,p38,and4eBP1signalinginresponsetoinfectionascomparedwithGr‐1−Tcellreceptorγδcells.Takentogether,Gr‐1expressionpatternsonγδTcellsinthelungprovidearobustmarkertodifferentiateinterferon‐γ–andinterleukin‐17A–producingsubsetsinvolvedintheearlyimmuneresponsetobacterialpneumonia...
