Frequencies of FoxP3+ naïve T cells are related to both viral load and naïve T cell proliferation responses in HIV disease

HIVinfectionresultsindepletionanddysfunctionofnaïveCD4+Tcells.Themechanismsunderlyingthesedeficienciesarenotunderstood.WeinvestigatedthefrequenciesofCD4+naïvesubsetsinHIVdiseaseasdefinedbyexpressionofCD25and/orFoxP3andtherelationshipofthesefrequenciestonaïveTcellproliferationfunction.WeobservedincreasedproportionsofCD25+FoxP3+andCD25+FoxP3–cellsanddecreasedproportionsofCD25–FoxP3–cellswithinthenaïveCD4+cellcompartmentfromHIV‐infectedpersonscomparedwithfindingsinhealthydonors.TheseperturbationswererelatedtohigherplasmaHIVRNAlevelsbutnotwithhigherimmuneactivation,asmeasuredbytheproportionsofCD38+memoryCD4+Tcells.NaïveTcellproliferationresponsestomitogenstimulationwereinverselyrelatedtothefrequenciesandabsolutenumbersofFoxP3+naïveTcells.MDA,amarkerofoxidativestress,andsCD14,amarkerofmonocyteactivationandasurrogateformicrobialtranslocation,wereincreasedinserumsamplesfromHIV+donors;however,neithermarkerwasrelatedtonaïveTcellfunctioninHIV+donors.TheseobservationssuggestthatalterationsinnaïveTcellsubsetfrequenciescouldcontributetonaïveTcelldysfunctioninHIVdisease,butthesealterationsarenotnecessarilytheresultofchronicimmuneactivation...
