Amongthemechanismssetinmotionbythetumortoescapethecontroloftheimmunesystem,MDSCsplayacentralroleininducingtolerancetoavarietyofanti‐tumoreffectors,includingTlymphocytes.IthasbeendemonstratedthatMDSCsexpandintumor‐bearingmiceandincancerpatients,leadingtoanimpairmentofTcellreactivityagainstthetumor.However,asthepresenceofMDSCsisnotcorrelatedwithageneralimmunesuppression,itwasadvancedthatamechanismregulatingthespecificityofMDSCinhibitionmustbepresent.Inthisarticle,wereviewtheliteratureshowingthatMDSCsexerttheirimmune‐suppressivefunctiononAg‐specificTcellresponsesbutattimes,alsoonmitogen‐activatedTlymphocytes,thereforebypassingtheAgdependency.WeproposethatthefeaturesofMDSC‐mediatedimmunesuppressionmightbeinfluencednotonlybythespecificmicroenvironmentinwhichMDSCsexpandandbythetumorcharacteristicsbutalsobythelevelsofactivationofthetargetlymphocytes...
