Th1/Th2cytokinesplayakeyroleinimmuneresponsestoLeishmaniamajorbycontrollingmacrophageactivationforNOproductionandparasitekilling.MDSCs,includingmyeloidprecursorsandimmaturemonocytes,produceNOandsuppressTcellresponsesintumorimmunity.WehypothesizedthatNO‐producingMDSCscouldhelpimmunitytoL.majorinfection.Gr1hi(Ly6Chi)CD11bhiMDSCselicitedbyL.majorinfectionsuppressedpolyclonalandantigen‐specificTcellproliferation.Moreover,L.major‐inducedMDSCskilledintracellularparasitesinaNO‐dependentmannerandreducedparasiteburdeninvivo.Bycontrast,treatmentwithATRA,whichinducesMDSCstodifferentiateintomacrophages,increaseddevelopmentoflesions,parasiteload,andTcellproliferationindrainingLNs.Altogether,theseresultsindicatethatNO‐producingMDSCshelpprotectiveimmunitytoL.majorinfection,despitesuppressedTcellproliferation...
