Theβ2‐integrinsareasubfamilyofintegrinsexpressedonleukocytesthatplayanessentialroleinleukocytetrafficking,activation,andmanyotherfunctions.StudiesinEAE,theanimalmodelformultiplesclerosis,showdifferentialrequirementsforβ2‐integrinsinthisdiseasemodel,rangingfromcriticalinthecaseofLFA‐1(CD11a/CD18)tounimportantinthecaseofCD11d/CD18.Importantly,expressionofβ2‐integrinsonTcellsubsetsprovidessomecluesastothefunction(s)theseadhesionmoleculesplayindiseasedevelopment.Forexample,transferredEAEstudieshaveshownthatMac‐1(CD11b/CD18)expressiononαβTcellsiscriticalfordiseasedevelopment,andtheabsenceofLFA‐1onTregsinrecipientmiceresultsinexacerbateddisease.Inthisreview,wesummarizerecentfindingsregardingtheroleofβ2‐integrinsindemyelinatingdiseaseandnewinformationabouttheroleofβ2‐integrinswithrespecttoalterationsinTregnumbersandfunction.Inaddition,wediscussthepotentialfortargetingβ2‐integrinsinhumandemyelinatingdiseaseinlightoftherecentanimalmodelstudies...
