Monocytesandmacrophagesareadiversepopulationofinnateimmunecellsthatplayacriticalroleinhomeostasisandinflammation.Thesecellsaresurveillantbynatureandcloselymonitorthevasculatureandsurroundingtissueduringstatesofhealthanddisease.Giventheirabundanceandstrategicpositioningthroughoutthebody,myeloidcellsareamongthefirstresponderstoanyinflammatorychallengeandareactiveparticipantsinmostimmune‐mediateddiseases.Recentstudieshaveshednewlightonmyeloidcelldynamicsandfunctionbyuseofanimagingtechniquereferredtoasintravitalmicroscopy(IVM).Thispowerfulapproachallowsresearcherstogainreal‐timeinsightsintomonocytesandmacrophagesperforminghomeostaticandinflammatorytasksinlivingtissues.Inthisreview,wewillpresentacontemporarysynopsisofhowintravitalmicroscopyhasrevolutionizedourunderstandingofmyeloidcellcontributionstovascularmaintenance,microbialdefense,autoimmunity,tumorigenesis,andacute/chronicinflammatorydiseases...
