Manypriondiseasesareacquiredbyperipheralexposure,andskinlesionsareaneffectiverouteoftransmission.Followingexposure,earlyprionreplication,uponFDCsinthedrainingLNisobligatoryforthespreadofdiseasetothebrain.However,themechanismbywhichprionsareconveyedtothedrainingLNisuncertain.Here,transgenicmicewereused,inwhichlangerin+cells,includingepidermalLCsandlangerin+classicalDCs,werespecificallydepleted.Thesewereusedinparallelwithtransgenicmice,inwhichnonepidermalCD11c+cellswerespecificallydepleted.Ourdatashowthatprionpathogenesis,followingexposureviaskinscarification,occurredindependentlyofLCandotherlangerin+cells.However,thedepletionofnonepidermalCD11c+cellsimpairedtheearlyaccumulationofprionsinthedrainingLN,implyingaroleforthesecellsinthepropagationofprionsfromtheskin.Therefore,together,thesedatasuggestthatthepropagationofprionsfromtheskintothedrainingLNoccursviadermalclassicalDCs,independentlyoflangerin+cells...
