ThedeciduabasalisofdevelopingmouseimplantationsitesishighlyenrichedinCD45+leukocytes.Inintact,syngeneicallymatedC57BL/6deciduabasalisexaminedatgestationday8.5bywhole‐mountinsituimmunohistochemistry,leukocyte,butnottrophoblast,conjugationswerereported.Nothingisknownregardingtimecourse,frequency,composition,orimportanceofphysiologicdecidualCD45+cellpairing.Inthisstudy,weconfirmedthepresenceofanti‐CD54+/anti‐CD11a+immunesynapsesinCD45+decidualcellconjugatesandcharacterizedtheircellularheterogeneity.Conjugatedcellpairswerevirtuallyabsentbeforeimplantation(virginandgestationdays3.5and4.5),wereinfrequentatgestationday5.5,butinvolved19%ofallCD45+cellsbygestationday8.5,thendeclined.Bygestationday8.5,almostallCD45+cellscoexpressedCD31,and2CD45+CD31+cellscomposedmostconjugates.Conjugationpartnersweredefinedfor2nonoverlappinguterinenaturalkillercellsubsets(Ly49C/I+/Dolichosbiflorusagglutininlectin−andLy49C/I−/Dolichosbiflorusagglutininlectin+).Ly49C/I+uterinenaturalkillercellswerethemajorsubsetfrombeforematinguptogestationday6.5.Atgestationday5.5/6.5,uterinenaturalkillercellconjugatesinvolvingLy49C/I+cellsweremoreabundant.Bygestationday8.5/9.5,Dolichosbiflorusagglutininlectin+uterinenaturalkillercellswerethedominantsubsetwithDolichosbiflorusagglutininlectin+/Dolichosbiflorusagglutininlectin+homologousconjugatesandDolichosbiflorusagglutininlectin+/Dolichosbiflorusagglutininlectin−heterologousconjugatesdominatinguterinenaturalkillercellpairings.Atgestationday6.5,bothLy49C/I+/CD45+andDolichosbiflorusagglutininlectin+/CD45+heterologousconjugatepairsstronglyengagedantigen‐presentingcells(CD11c+,CD68+,ormajorhistocompatibilitycomplexclassII+).Bygestationday8.5,dominantpartnersofLy49C/I+/CD45+andDolichosbiflorusagglutininlectin+/CD45+heterologousconjugatesareTcells(CD8+>CD4+).HeterologousconjugatesthatdidnotinvolveuterinenaturalkillercellsoccurredbutdidnotsuggestantigenpresentationtoTcells.Thesedataidentifygestationday6.5–8.5inthepregnantmouseasacriticalwindowforleukocyteinteractionsthatmayestablishimmuneregulationwithinimplantationsites...
