LittleisknownofhowastrongimmuneresponseinthelungsisregulatedtominimizetissueinjuryduringsevereinfluenzaAvirus(IAV)infection.Here,usingamodeloflethal,high‐pathogenicityIAVinfection,wefirstshowthatLy6ChiLy6G–inflammatorymonocytes,andnotneutrophils,arethemaininfiltrateinlungsofWTmice.MicedevoidofiNKTcells(Jα18−/−mice)haveincreasedlevelsofinflammatorymonocytes,whichcorrelatedwithincreasedlunginjuryandmortality(butnotviralload).ActivationofiNKTcellscorrelatedwithreductionofMCP‐1levelsandimprovedoutcome.iNKTcellswereabletoselectivelylyseinfected,MCP‐1‐producingmonocytesinvitro,inaCD1d‐dependentprocess.OurstudyprovidesadetailedprofileandkineticsofinnateimmunecellsinthelungsduringsevereIAVinfection,highlightinginflammatorymonocytesasthemajorinfiltrateandidentifyingaroleforiNKTcellsincontrolofthesecellsandlungimmune‐pathology...
