Comparedwiththestablecoretemperature,theskintemperatureislowerandvariesdependingonambienttemperatureandconvectionconditions.ThefunctionofDCs,whichareplentifulintheskinatlowerphysiologicaltemperatures,hasnotbeenreported.WeshowthatDCperformedsomefunctionsnormallyat28°C,includingphagocytosisandmacropinocytosis.TLR‐4signalingviaMAPKpathwayswasdelayedat28°Cbutreachednormallevels,whichmayexplaintheobservedslowerkineticsofstimulatedmacropinocytosisandTNFproduction.TLR‐4‐inducedNOproductionwascompromisedseverelyat28°C.Collagendegradationandmigrationthroughmatrigel‐coatedtranswellinsertsweredecreased,butnoeffectonpodosomenumberorDCmigrationthroughnoncoatedtranswellfilterswasseen.LoweringthetemperaturedifferentiallyregulatedfunctionsassociatedwiththeroleofDCsinadaptiveimmunity.LPS‐inducedup‐regulationofCD86wasnormal;however,CD40up‐regulationwassuppressedafterTLR‐4stimulationat28°C.NonactivatedDCprocessedandpresentedantigenonMHCclassIIequallywellat28°Cand37°C.However,DCsthatwereloadedwithantigensandstimulatedwithTLRligandat28°CwerepooratactivatingTcellsat37°CcomparedwithDCsthatwereactivatedandloadedwithantigenat37°C...
