ShortpeptidefragmentsgeneratedbyintracellularproteincleavagearepresentedonthesurfaceofmostnucleatedcellsboundtohighlypolymorphicMHCImolecules.ThesepMHCIcomplexesconstituteaninterfacethatallowstheimmunesystemtoidentifyanderadicateanomalouscells,suchasthosethatharborinfectiousagents,throughtheactivationofCTLs.MolecularrecognitionofpMHCIcomplexesismediatedprimarilybyclonallydistributedTCRsexpressedonthesurfaceofCTLs.ThecoreceptorCD8contributestothisantigen‐recognitionprocessbybindingtoalargelyinvariantregionoftheMHCImoleculeandbypromotingintracellularsignaling,theeffectsofwhichservetoenhanceTCRstimulitriggeredbycognateligands.RecentinvestigationshaveshedlightontheroleofCD8intheactivationofMHCI‐restricted,antigen‐experiencedTcellsandintheprocessesofTcellselectionandlineagecommitmentinthethymus.Here,wereviewthesedataanddiscusstheirimplicationsforthedevelopmentofpotentialtherapeuticstrategiesthatselectivelytargetpathogenicCTLresponseserroneouslydirectedagainstself‐derivedantigens...
