Arapidandrobustrecruitmentofcirculatingneutrophilsatsitesofinfectioniscriticalforpreventingbacterialspread.Theefficiencyofthisprocess,however,isgreatlydiminishedduringsepsis,aseveresystemicinflammatoryresponsetoinfection.Theproteolyticactivityofadisintegrinandmetalloprotease‐17isinducedinthecellmembraneofleukocytesupontheiractivation,resultingintheconversionofmembranetosolubleTNF‐αandthereleaseofassortedreceptorsfromthesurfaceofneutrophilsimportantfortheireffectorfunctions.Weshowthatconditionalknockoutmicelackingadisintegrinandmetalloprotease‐17inallleukocyteshadasurvivaladvantagewhensubjectedtopolymicrobialsepsis.Bacteremiaandthelevelsofcirculatingproinflammatorycytokines,keydeterminantsofsepsisseverity,weresignificantlyreducedinconditionaladisintegrinandmetalloprotease‐17knockoutmiceduringsepsis.Althoughcecalbacterialmicrobiotaandloadweresimilarinunmanipulatedconditionaladisintegrinandmetalloprotease‐17knockoutandcontrolmice,peritonealspreadofbacteriawassignificantlyreducedinconditionaladisintegrinandmetalloprotease‐17knockoutmicefollowingsepsisinduction,whichwasassociatedwithanamplifiedrecruitmentofneutrophils.Takentogether,ourfindingssuggestthatextensiveadisintegrinandmetalloprotease‐17inductionduringsepsismaytipthebalancebetweenefficientandimpairedneutrophilrecruitment...
