CellularAFisusuallyconsideredahindrancetoflowcytometricanalysis.Here,weincorporateAFintoanalysisofcomplexmixturesofleukocytes.Usingamousemodel,weexaminedcellularAFatmultipleexcitationandemissionwavelengths,andpopulationswithdiscretepatternsweregatedandexaminedforsurfacemarkerexpression.Inthespleen,allmajormyeloidpopulationswereidentified.Inparticular,theapproachallowedsimultaneouscharacterizationofRPMandresidentmonocytes.WhenmonocytesandRPMwerecompared,RPMexhibitedaphenotypethatwasconsistentwithinvolvementinphysiologicalprocesses,includingexpressionofgenesinvolvedinlipidandironmetabolism.ThepresenceoflargeamountsofstoredferricironwithinRPMenabledpurificationofthesecellsusingamagnetic‐basedapproach.Whenadaptedforuseonleukocytesisolatedfromarangeofotherorgans,incorporationofAFintoanalysisallowedidentificationandisolationofbiologicallyimportantmyeloidpopulations,includingsubsetsthatwerenotreadilyidentifiablebyconventionalcytometricanalysis...
