CX3CL1,orfractalkine,theuniquememberoftheCX3Cchemokinefamily,existsasatransmembraneglycoprotein,aswellasinsolubleform,eachmediatingdifferentbiologicalactivities,andisconstitutivelyexpressedinmanyhematopoieticandnonhematopoietictissues.CX3CR1,theCX3CL1exclusivereceptor,isaclassicalGPCR,expressedonNKcells,CD14+monocytes,andsomesubpopulationofTcells,Bcells,andmastcells.ArecentpaperbyourgrouphasdemonstratedforthefirsttimethathighlypurifiedhumanBcellsfromtonsilandperipheralbloodexpressedCX3CR1atmRNAandproteinlevels.Inparticular,tonsilnaïve,GC,andmemoryBcellsexpressedCX3CR1,butonlyGCcentrocyteswereattractedbysolubleCX3CL1,whichwithitsreceptor,arealsoinvolvedinthepathogenesisofseveralinflammatorydisorders,aswellasofcancer.PreviousstudieshaveshownthatCX3CR1isup‐regulatedindifferenttypesofBcelllymphoma,aswellasinB‐CLL.Recently,wehavedemonstratedthattheCX3CL1/CX3CR1axisisinvolvedintheinteractionofB‐CLLcellswiththeirmicroenvironment.Takentogether,ourdatadelineateanovelrolefortheCX3CL1/CX3CR1complexinthebiologyofnormalBcellsandB‐CLLcells.Thesetopicsarethesubjectofthisreviewarticle...
