CA‐MRSAinfectionsareoftencausedbystrainsencodingPVL,whichcancauselysisofPMNsandothermyeloidcellsinvitro,afunctionconsideredwidelyastheprimarymeansbywhichPVLmightcontributetodisease.However,atsublyticconcentrations,PVLcanfunctionasaPMNagonist.Tobetterunderstandthisphenomenon,weinvestigatedtheabilityofPVLtoalterhumanPMNfunction.PMNsexposedtoPVLhadenhancedcapacitytoproduceO2−inresponsetofMLF,butunlikeprimingbyLPS,thisresponsedidnotrequireTLRsignaltransduction.Ontheotherhand,therewassubcellularredistributionofNADPHoxidasecomponentsinPMNsfollowingexposureofthesecellstoPVL—afindingconsistentwithpriming.Importantly,PMNsprimedwithPVLhadanenhancedabilitytobind/ingestandkillStaphylococcusaureus.PrimingofPMNswithotheragonists,suchasIL‐8orGM‐CSF,alteredtheabilityofPVLtocauseformationofporesintheplasmamembranesofthesecells.MicroarrayanalysisrevealedsignificantchangesinthehumanPMNtranscriptomefollowingexposuretoPVL,includingup‐regulationofmoleculesthatregulatetheinflammatoryresponse.Consistentwiththemicroarraydata,mediatorsoftheinflammatoryresponsewerereleasedfromPMNsafterstimulationwithPVL.WeconcludethatexposureofhumanPMNstosublyticconcentrationsofPVLelicitsaproinflammatoryresponsethatisregulatedinpartatthelevelofgeneexpression.WeproposethatPVL‐mediatedprimingofPMNsenhancesthehostinnateimmuneresponse...
