Osteoclastsarehighlyspecializedcellsthatresorbboneandcontributetoboneremodeling.Diseasessuchasosteoporosisandosteolyticbonemetastasisoccurwhenosteoclast‐mediatedboneresorptiontakesplaceintheabsenceofconcurrentbonesynthesis.Considerableefforthasbeenplacedonidentifyingmoleculesthatregulatetheboneresorptionactivityofosteoclasts.Tothisend,weinvestigateduniqueandoverlappingfunctionsofmembersoftheFAKfamily(FAKandPyk2)inosteoclastfunctions.Withtheuseofaconditionalknockoutmousemodel,inwhichFAKisselectivelytargetedfordeletioninosteoclastprecursors(FAKΔmyeloid),wefoundthatlossofFAKresultedinreducedboneresorptionbyosteoclastsinvitro,coincidentwithimpairedsignalingthroughtheCSF‐1R.However,bonearchitectureappearednormalinFAKΔmyeloidmice,suggestingthatPyk2mightfunctionallycompensateforreducedFAKlevelsinvivo.Thiswassupportedbydatashowingthatpodosomeadhesionstructures,whichareessentialforbonedegradation,weresignificantlymoreimpairedinosteoclastswhenFAKandPyk2werereducedthanwheneithermoleculewasdepletedindividually.WeconcludethatFAKcontributestocytokinesignalingandboneresorptioninosteoclastsandpartiallycompensatesfortheabsenceofPyk2tomaintainproperadhesionstructuresinthesecells...
