Successfulimmunedefenserequiresintegrationofmultipleeffectorsystemstomatchthediversevirulencepropertiesthatmembersofthemicrobialworldmightexpressastheyinitiateandpromoteinfection.Humanneutrophils—thefirstcellularresponderstoinvadingmicrobes—exertmostoftheirantimicrobialactivityinphagosomes,specializedmembrane‐boundintracellularcompartmentsformedbyingestionofmicroorganisms.ThetoxinsgenerateddenovobythephagocyteNADPHoxidaseanddeliveredbyfusionofneutrophilgranuleswithnascentphagosomescreateconditionsthatkillanddegradeingestedmicrobes.Antimicrobialactivityreflectsmultipleandcomplexsynergiesamongthephagosomalcontents,andoptimalactionreliesonoxidantsgeneratedinthepresenceofMPO.Theabsenceoflife‐threateninginfectiouscomplicationsinindividualswithMPOdeficiencyisfrequentlyofferedasevidencethattheMPOoxidantsystemisancillaryratherthanessentialforneutrophil‐mediatedantimicrobialactivity.However,thatargumentfailstoconsiderobservationsfromhumansandKOmicethatdemonstratethatmicrobialkillingbyMPO‐deficientcellsislessefficientthanthatofnormalneutrophils.WepresentevidenceinsupportofMPOasamajorarmofoxidativekillingbyneutrophilsandproposethattheessentialcontributionofMPOtonormalinnatehostdefenseismanifestonlywhenexposuretopathogensoverwhelmsthecapacityofotherhostdefensemechanisms...
