ThismanuscriptdescribestechnicaladvancesallowingmanipulationandquantitativeanalysesofhumanDCmigratorybehaviorinlungepithelialtissue.DCsarehematopoieticcellsessentialforthemaintenanceoftissuehomeostasisandtheinductionoftissue‐specificimmuneresponses.Importantfunctionsincludecytokineproductionandmigrationinresponsetoinfectionfortheinductionofproperimmuneresponses.TodesignappropriatestrategiestoexploithumanDCfunctionalpropertiesinlungtissueforthepurposeofclinicalevaluation,e.g.,candidatevaccinationandimmunotherapystrategies,wehavedevelopedalive‐imagingassaybasedonourpreviouslydescribedorganotypicmodelofthehumanlung.ThisassayallowsprovocationsandsubsequentquantitativeinvestigationsofDCfunctionalpropertiesunderconditionsmimickingmorphologicalandfunctionalfeaturesoftheinvivoparentaltissue.Wepresentprotocolstosetupandpreparetissuemodelsfor4D(x,y,z,time)fluorescence‐imaginganalysisthatallowspatialandtemporalstudiesofhumanDCsinliveepithelialtissue,followedbyflowcytometryanalysisofDCsretrievedfromdigestedtissuemodels.ThismodelsystemcanbeusefulforelucidatingincompletelydefinedpathwayscontrollingDCfunctionalresponsestoinfectionandinflammationinlungepithelialtissue,aswellastheefficacyoflocallyadministeredcandidateinterventions...
