【 摘 要 】
CathelicidinLL‐37isamultifunctional,immunomodulatoryandantimicrobialhost‐defensepeptideofthehumanimmunesystem.Here,weidentifiedtheroleofSFKsinmediatingthechemokineinductionactivityofLL‐37inmonocyticcells.LL‐37inducedSFKphosphorylation;andchemicalinhibitorsofSFKssuppressedchemokineproductioninresponsetoLL‐37stimulation.SFKswererequiredforthedownstreamactivationofAKT,butCa2+‐fluxandMAPKinductionwereSFK‐independent.ThroughsystematicsiRNAknockdownofSFKmembers,arequirementforLyninmediatingLL‐37activitywasidentified.TheinvolvementofLynincathelicidinactivitieswasfurtherconfirmedusingLyn‐knockoutmouseBMDMs.TheroleofSFKsandLynwasalsodemonstratedintheactivitiesofthesyntheticcationicIDRpeptides,developedasnovel,immunomodulatorytherapeutics.ThesefindingselucidatethecommonmolecularmechanismsmediatingthechemokineinductionactivityofnaturalandsyntheticcationicpeptidesinmonocyticcellsandidentifySFKsasapotentialtargetformodulatingpeptideresponses...
