【 摘 要 】
Lipopolysaccharide(LPS)isapotentnaturaladjuvant,commonlyusedtoamplifyTh1responses.Here,wereportthatsystemicimmunizationusingLPSgenerateslargenumbersofspecificTh17cellsinmurinesmallintestinallaminapropria.TheprimingoftheseTh17cellsrequiredIL‐23p19productionbybonemarrow‐derivedcells.Incontrast,IL‐23hadnoimpactonTh1differentiationoroverallnumbersofAg‐specificregulatoryTcells.ExperimentsusingT‐celladoptivetransfersrevealedapreviouslyunappreciatedmechanismforhowTh17responsesareamplifiedinvivo:stimulationthroughLPSexpandedprecommittedTh17cellsratherthancausingTh17differentiation.Second,LPSdroveTh17cellexpansionindependentlyofIL‐23,demonstratingthatthiscytokineisnotnecessaryforexpansionandpossiblyfunctionsatanearlierstageinTh17priming.OurdataprovideanimpetusforusingLPS‐basedperipheralvaccinationtoaugmentspecificT‐cell‐mediatedimmunityinthegutmucosa...
