【 摘 要 】

Tregsplayimportantrolesinmaintainingimmunehomeostasis,andthus,therapiesbasedonTregarepromisingcandidatesforthetreatmentforavarietyofimmune‐mediateddisorders.Thesetherapies,however,facethesignificantchallengeofobtainingadequatenumbersofTregsfromperipheralbloodthatmaintainssuppressivefunctionfollowingextensiveexpansion.InducingTregsfromnon‐Tregsoffersaviablealternative.DifferentmethodstoinduceTregshavebeenproposedandinvolvemainlytreatingcellswithTGF‐β‐iTreg.However,useofTGF‐βaloneisnotsufficienttoinducestableTregs.ATRAorrapahasbeenshowntosynergizewithTGF‐βtoinducestableTregs.WhereasTGF‐βplusRA‐iTregshavebeenwell‐describedintheliterature,thephenotype,function,andmigratorycharacteristicsofTGF‐βplusrapa‐iTreghaveyettobeelucidated.Herein,wedescribethephenotypeandfunctionofmouserapa‐iTregandrevealthatthesecellsdifferintheirinvivohomingcapacitywhencomparedwithmouseRA‐iTregandmouseTGF‐β‐iTreg.Thisdifferenceinmigratoryactivitysignificantlyaffectsthetherapeuticcapacityofeachsubsetinamousemodelofcolitis.WealsodescribethecharacteristicsofiTreggeneratedinthepresenceofTGF‐β,RA,andrapa...
