【 摘 要 】

Theuseoflive,attenuated,orgeneticallymodifiedmicrobesortheircellularcomponent(s)ormetaboliteshasbeguntoemergeasapotentialnewapproachinmedicinalresearchtodeliverbiologicallyactiveentities.Thus,advancingourknowledgeofsuchmicrobe‐mediatedtherapymaysuggestnewavenuesfortherapeuticinterventioninmanydiseases.WehadearlierreportedthatthetotallipidofattenuatedLeishmaniadonovanisuppressedtheinflammatoryresponsesinrheumatoidarthritispatients.OurpresentstudyrevealsthatthepLLD,isolatedfrompathogenicL.donovani,decreasestheinflammatorylevelofbacterialendotoxininstimulatedmousemacrophages,asalsointheinvivomurinesystem.Itexertstheactivitybyreducingthelevelofdifferentmediators,suchascytokine‐chemokine(s).ItalsosuppressestheexpressionoftheubiquitoustranscriptionfactorNF‐κBp65instimulatedmacrophagecells,improvestheendotoxin‐associatedliverdamage,reducesthevascularpermeabilityfactors,suchasVEGF,andsuppressestheexpressionofcelladhesionmolecules,includingICAM‐1,VCAM‐1,PECAM‐1,P‐selectin,andE‐selectin,inliverofsepticmice.ThesefindingsindicatethatpLLDmayprovetobeapotentialanti‐inflammatoryagentandprotectfromendotoxin‐inducedsepsisinhepaticimpairment...
