AscreenofcellsurfacemarkersdifferentiallyexpressedduringperipheralBcelldifferentiationidentifiedthattheCD45RBepitopedetectedbythemAbMEM‐55washighlyexpressedonCD27+memoryBcellsandabsentonCD27–naïveBcells.IgG+CD27–memoryandapreviouslyunacknowledgedCD27–populationinbloodalsoexpressedhighlevelsofCD45RBMEM55.NaïveandmemoryBcellsfromtonsilsfollowedthepatternobservedinblood,andCD38highBcellshadabimodalexpressionpatternwhenanalyzedusingflowcytometry.NoCD38highGCBcells,however,expressedtheCD45RBMEM55epitopewhenassayedusingimmunohistochemistry.Rather,CD38highCD45RBMEM55highBcellshadadistinctcellularphenotypeandwerelocalizedoutsideofGCs.CD45RBepitopes,detectedbyotherantibodyclones,wereexpressedathighlevelsthroughBcelldifferentiation,andnochangesinsplicingoftheCD45RBexonwereobservedduringBcelldifferentiation.Instead,BcellsregulatedtheirexpressionoftheCD45RBMEM55epitopethroughsite‐specificmodificationsofanO‐linkedglycochain.CD4+TcellsdifferentiallysplicedCD45butdidnotvarytheglycosylationoftheCD45RBMEM55epitope,andCD8+cellsmodifiedCD45RBMEM55expressioninasimilarmannerasBcells.MonocytesexpressedtheCD45RBexonbutnottheCD45RBMEM55epitope.AsCD45isahighlyexpressedtyrosinephosphatasethatregulatesantigenreceptorsignalingstrengthinlymphocytes,weconcludethatregulatedO‐linkedglycosylationofCD45RBcanbeusedtofollowBcelldifferentiationandthatthisregulationmaybeinvolvedinfine‐tuningantigensignalinginthecell...
