【 摘 要 】
MastcellsarecrucialinthedevelopmentofimmunityagainstBordetellapertussis,andthefunctionofTLRsinthisprocesshasbeeninvestigated.Here,theinteractionbetweenmastcellsandB.pertussiswithanemphasisontheroleofCLRsisexamined.Inthisstudy,twoCLRs,MGLandMR,weredetectedforthefirsttimeonthesurfaceofmastcells.TheinvolvementofMRandMGLinthestimulationofmastcellsbyheat‐inactivatedBPwasinvestigatedbytheuseofblockingantibodiesandspecificcarbohydrateligands.ThecellwallLOSofBPwasalsoisolatedtoexploreitsroleinthisinteraction.Mastcellsstimulatedwithheat‐inactivatedBPorBPLOSinducedTNF‐α,IL‐6,andIFN‐γsecretion,whichwassuppressedbyblockingMRorMGL.InhibitionofCLRssignalingduringBPstimulationaffectedtheabilityofmastcellstopromotecytokinesecretioninTcellsbuthadnoeffectonthecell‐surfaceexpressionofICAM1.BlockingMRorMGLsuppressedBP‐inducedNF‐κBexpressionbutnotERKphosphorylation.SykwasinvolvedintheCLR‐mediatedactivationofmastcellsbyBP.BacterialrecognitionbyimmunecellshasbeenpredominantlyattributedtoTLRs;inthisstudy,thenovelroleofCLRsintheBP–mastcellinteractionishighlighted...
